Larval brains expressing dpnScer\UAS.cSa under the control of Scer\GAL4insc-Mz1407 exhibit increased numbers of type II neuroblasts, as compared to controls.

The brains of third instar larvae expressing dpnScer\UAS.cSa under the control of either Scer\GAL4wor.PA or Scer\GAL4elav.PU show an increased number of mira positive neuroblasts. The neuroblasts are found deep within the brain forming a compact mass. The increase in neuroblasts is at the expense of GMCs and neurons. The lack of ase expression suggests that the extra neuroblasts seen are type II neuroblasts.

When dpnScer\UAS.cSa is expressed under the control of Scer\GAL4elav.PU an increase in brain size is seen at the end of larval development. The effects are much weaker in the ventral part of the brain compared to the dorsal region. No extra neuroblasts are seen when dpnScer\UAS.cSa is expressed under the control of Scer\GAL4ase.PU.

Type I neuroblast clones expressing dpnScer\UAS.cSa under the control of Scer\GAL4αTub84B.PL are indistinguishable from control clones. Clones induced in type II neuroblasts are comprised mostly of cells that express markers consistent with either type II neuroblasts of immature INP. The small size of these cells is similar to INPs, but unlike INPS they are mitotically active.

Co-expression of bansponge.Scer\UAS.T:Disc\RFP leads to partial suppression of the increased neuroblast numbers seen in larval brains expressing dpnScer\UAS.cSa under the control of Scer\GAL4insc-Mz1407.

Expressing dpnScer\UAS.cSa in clones under the control of Scer\GAL4Scer\FRT.Act5C suppresses neuroblast loss seen when Nl1N-ts1 is shifted to the restrictive temperature from the first instar larval stage onwards.
