An attP attachment site is first targeted distal to the most distal essential gene mod on chromosome 3R via homologous recombination. A fragment consisting of a 240-copy tandem repeat of lacO sites together with a proximal y marker and a distal I-SceI recognition site is integrated at the attP site next. Subsequently, sequences distal to the I-SceI recognition site including Map205 are deleted creating an artificial telomere marked with an lacO-array.
