Egg chambers arrest at stage S10. The oldest egg chambers often contain polytene nurse cell nuclear chromatin with high ploidy. cup4 has a dominant effect on meiotic X chromosome segregation.

Homozygous females show a strong phenotypic improvement if they also carry one copy of otu11, allowing egg chamber growth to a later developmental stage with more uniform chromatin morphologies, although they are still sterile. 41% of egg chambers in these females are dicephalic. Rbp9B/Rbp9B cup4/+ females are weakly fertile, with an average of one viable embryo per female.
