Description

Construct: The last 20bp of the tenth exon and the first 11bp of the 11th exon of the 4.8kb ninaC mRNA are fused together, deleting a 582bp intron and a 3' end processing signal. This specifically eliminates expression of the p132 ninaC protein.

Flies expressing ninaCΔ132 in a ninaC5 background have a slower rate of termination of the photoresponse (measured using an electroretinogram recording) than wild-type flies after pre-exposure to light for 10 minutes.

Photoreceptors treated with 8-Br-cAMP or IBMX show slowing of macroscopic light response, as do wild type, but unlike ninaC5 or ninaCΔ174.

Plateau response to steady light or flashing light is normal. Pupillary pigment migration is smaller than that in wild type.

The electroretinogram of ninaC5 flies carrying ninaCΔ132 does not differ with age or with rearing on a light/dark cycle, and no significant reduction in rhabdomeral diameter is seen after 21 days in a light/dark cycle.
