Scer\GAL4how-24B-mediated expression of UbxIa.Scer\UAS results in an ectopic DA3 muscle in thoracic segment 1. The number of nuclei in this ectopic muscle is identical to that in wild type abdominal segments.
Expression of UbxIa.Scer\UAS using Scer\GAL4kirre-rP298, but not Scer\GAL4sns.PK, converts the number of nuclei in thoracic segments 2 and 3 to that found in abdominal segments.
85% of the embryonic/first instar larval cuticles of UbxIa.Scer\UAS; Scer\GAL4arm.PS animals lack Keilin's organs.
Scer\GAL4444-mediated expression at 17oC causes the thoracic and some head segments of first instar larvae to display A1 identity. At 29oC larvae develop all segments anterior to A2 with an A3-A5 pattern.
When expressed via Scer\GAL41151, T2 myoblast migration during pupal development is unaffected. The indirect flight muscle fail to complete development and eventually degenerate.
Scer\GAL4E132 induced expression in the eye-antennal disc causes transformation of the antenna into leg. Scer\GAL4Bx-MS1096 or Scer\GAL4C-765 induced expression in the wing disc causes transformation of the wing into haltere-like tissue. Co-expression of P{UAS-exd.G} (induced by Scer\GAL4Bx-MS1096) does not significantly modify the wing transformation. Scer\GAL4C-765 induced expression also transforms the mesonotum into the homologous region of the third thoracic segment. Scer\GAL4c739 and Scer\GAL4C-765 induced expression in the abdomen fails to cause segmental change. Ubxabx-1 Ubxbx-3 Ubxpbx-1/Ubxlac1 combination transforms the metathorax into mesothorax (including transformation of halteres into wings), Scer\GAL4Bx-MS1096 induced expression allows full rescue.
When expressed via Scer\GAL4how-24B in embryos, an ectopic precursor for abdominal muscle 26 appears in the ventral region of T1-T3. Ventral longitudinal muscles of thoracic segments assume the identity of the abdominal segments. Abdominal segment 1 gains two muscles that it normally lacks: external ventral oblique muscles 17 and 25. Insertion sites of transformed muscles are sometimes aberrant. The thoracic cuticle remains unaffected. Both gastric caeca and the first midgut constriction are missing.
Phenotype can be partially reverted by adding an additional dose of exd+.
