Transient expression assays have been carried out using derivatives of the AdhGC-1 allele. These derivatives contain varying lengths of 5' and 3' genomic sequences flanking the site of the Adh+t3.2 insertion in AdhGC-1, together with the Adh+t3.2 insertion. These assays indicate that a silencer element is located at least 6.5kb upstream of the Adh+t3.2 insertion in AdhGC-1. A negative element is also located downstream of the Adh+t3.2 insertion in AdhGC-1. These regulatory elements together produce the insertion specific expression pattern seen for the AdhGC-1 allele.
