Description

Construct: The 5' regulatory region, first exon, first intron and translational start site of Act88F are fused upstream of cDNA/genomic sequences encoding an embryonic form of Mhc lacking the globular head region. The Mhc coding sequences extend from 45 amino acids upstream of the head-rod junction through to the C-terminus. Exon 15b is included and is prespliced at the 3' end such that exon 18 is excluded (this alters the amino acid sequence at the C-terminus so that it matches the embryonic isoform). A 1.5kb fragment from the 3' region of Mhc (which includes polyadenylation signals) is included downstream of the Mhc coding sequences.

Flies carrying MhcR21-1.Act88F show reduced flight ability compared to controls. The degree of flight impairment correlates well with the level of MhcR21-1.Act88F protein accumulation. Thick and thin filaments are present in pupal indirect flight muscles (IFMs) of flies carrying MhcR21-1.Act88F, but myofibrils are structurally defective. Sarcomeres form although frequent cracks are seen in the myofilament lattice and sarcomere length and width are more variable than wild type. There are defects in the packing of thick filaments and the myofibrils are frequently not round in cross-section. Average sarcomere length is significantly reduced compared to wild type. Myofibril diameter is increased by approximately 20%. The number of thick filaments/cross-sectional area is reduced by approximately 40%. Mhc10 flies expressing MhcR21-1.Act88F have hollow thick filaments and myofibril-like structures in the IFMs. Hexagonal packing of the thick filaments with arrays of six thin filaments surrounding each thick filament is seen, as in wild type. Less rescue is seen than with MhcR57-24.Act88F.
