Expression of Mef2III.Scer\UAS under the control of Scer\GAL4P0.5.Pdf causes long locomotor activity rhythms, with a period of 25.1 +/- 0.1 hours.

Expression of Mef2III.Scer\UAS under the control of Scer\GAL4tim.Scer\UAS causes long locomotor activity rhythms, with a period of 24.6 +/- 0.3 hours. 61.5% of the flies have a single rhythm, 23.1% of flies have a complex rhythm (classified as more than one rhythmic period detectable above the 95% significance line using Lomb-Scargle analysis of periodograms) and 15.4% are arrhythmic.

Dorsal closure fails in embryos expressing Mef2III.Scer\UAS under the control of Scer\GAL4da.G32. The cuticle is thin but has denticle belts, although their pattern is abnormal. Expression of Mef2III.Scer\UAS under the control of Scer\GAL4twi.PB results in embryos that are phenotypically wild type.

At 18oC, expression of Mef2III.Scer\UAS under the control of Scer\GAL4how-24B rescues midgut constrictions in 90% of Mef222-21 embryos. The pharyngeal muscles are wild-type in 50% of the embryos. A subset of muscles, including some dorsal (dorsal transverse muscle 1), some lateral (lateral longitudinal muscle 1, segment border muscle) and some ventral muscles (ventral transverse muscle 1, ventral acute muscle 1, ventral acute muscle 2) are rescued. Unfused myoblasts are seen in the majority of hemisegments. The heart shows a range of phenotypes in these embryos. At 25oC, expression of Mef2III.Scer\UAS under the control of Scer\GAL4how-24B rescues midgut constrictions in Mef222-21 embryos. The shape of the pharyngeal muscle cells is not wild type in 30% of embryos, while it is fully rescued in other embryos. The somatic musculature is almost rescued to wild type, although occasionally one or two muscles in a hemisegment are absent or have an abnormal shape. The variety of phenotypes seen in the heart is similar to that seen when Mef2III.Scer\UAS is expressed under the control of Scer\GAL4how-24B at 18oC. At 29oC, when Mef2III.Scer\UAS is expressed under the control of Scer\GAL4how-24B in Mef222-21 embryos there are no midgut constrictions. The pharyngeal muscles are completely rescued. The heart is disorganised posteriorly in some embryos. The somatic musculature is fully rescued; the pattern of myosin-expressing fibres is essentially wild type, although there are some unfused myoblasts in the majority of hemisegments.
