Selected as: a recessive mutation showing reduced red eye pigmentation and adenine auxotrophy. The initial strain carrying "ade5²" - strain "H23-14" was only partially responsive to an RNA dietary supplement. It appeared that there were two classes of flies in this strain, one carrying a mutation with reduced red eye pigmentation and adenine auxotrophy (ade5²) and the other carrying a mutation with normal red eye pigmentation and auxotrophy where only slowly developing survivors occur on an RNA supplement (H23-14⁴⁴). Two "ade5²" stocks, ade5²⁽⁷⁾ and ade5²⁽²⁵⁾ have been derived from the original "H23-14" strain and they most likely carry the same mutation.

Alleles can be placed in order on the basis of relative survival on standard medium (from least to most viable): ade5¹, ade4¹, ade5²⁽⁷⁾, <up>ade5⁵, ade5⁶, ade5⁸</up>.
