cv-cdsRNA.Scer\UAS expression under the control of Scer\GAL4cv-c-C5 alone (the intrinsic Gal4 driver of cv-cC5), Scer\GAL4104Y alone, Scer\GAL4GMR23E10 alone, or Scer\GAL4GMR23E10 in combination with Gal80[ts] (in order to restrict expression to adulthood), induces a significant decrease in the mean sleep time of adults reared under 12h light:12h dark cycles, as compared to controls. Expression of cv-cdsRNA.Scer\UAS under the control of Scer\GAL4GMR23E10 alone also results in adults presenting a significantly lower sleep recovery after overnight sleep deprivation and a significant decrease in short-term olfactory memory under ad libitum sleep conditions, assessed by aversive tests to the 4-methylcyclohexanol odorant after electric shock-coupled training; these individuals, however present no significant changes in the sensitivity to electric shocks, as assessed by maximal motor velocities, and no changes in the preference score of naive individuals to the 4-methylcyclohexanol odorant in ad libitum sleep conditions, as compared to controls.
