Description

P-element sequences present in the P{3'lacW.2Bab-10}br2Bab-10 insertion (donor element) have replaced the defective P-element sequences present in the P{}br28 insertion (target element) in the br Z2 region. The converted transposon (P{lacW.2Bbc-1}) present in the P{lacW.2Bbc-1}br2Bbc-1 insertion is a chimera of the donor and target elements; at the 5' end, at least the first 33bp of the converted transposon is derived from the target element rather than the donor element, while at the 3' end, the converted transposon contains 344bp of P-element sequence (compared to 223bp present in the donor element) joined to the neo sequence at position 10385 of the published P{lacW} sequence, which indicates that 82bp of the neo sequence is missing (this deletion of neo sequences is not found in the donor element).

The puffing pattern of salivary gland polytene chromosomes in mutant larvae appears normal up to PS6-7 only; early puffs appear and disappear, intermolt puffs regress, mid-late puffs have a normal size, but late-late puffs are usually underdeveloped or do not appear at all. Just prior to the death of the larvae, the puffing pattern corresponds roughly to PS8-9 of normal development.
