50% of nesdEY08338 mutant males are sterile. Males exhibit multinucleate spermatids in 21% of cells. These spermatids display a failure of cytokinesis during one or both meiotic divisions. Onion stage spermatids contain two or four haploid nuclei accompanied by an enlarged Nebenkern.

The contractile ring constricts asymmetrically in 46% of nesdEY08338 mutant spermatocytes. Subsequently, the cleavage furrow collapses and the contractile ring fails to form a robust circular structure. Membrane invagination collapses and isolated contractile ring remnants are observed in the cytoplasm. Cytokinesis fails in 49% of spermatocytes, and plasma membrane invagination is asymmetric and incomplete in 27% of mutant cells. Ring canals fail to assemble during late cytokinesis.

80% of nesdEY08338/Df(2L)ED1305 mutant males are sterile. Males exhibit multinucleate spermatids in 32% of cells. These spermatids display a failure of cytokinesis during one or both meiotic divisions. Onion stage spermatids contain two or four haploid nuclei accompanied by an enlarged Nebenkern.

38% of nesdEY08338 mutant egg chambers at stage 9 and 10 contain remnants of ring canals associated with binucleate nurse cells, which is never observed in control animals or nesdEY08338 egg chambers from an earlier developmental stage.

Expression of pavUbi-p63E.T:Avic\GFP-m6 fully rescues the cytokinesis and sterility defects of nesdEY08338/Df(2L)ED1305 mutant flies, but not the lethality of the mutants.

nesdEY08338/Df(2L)ED1305 mutant flies with one copy of pavB200 show a two-fold increase in then number of multinucleate spermatids.
