In wild type pupal notum epithelium cells, microtubule ablation causes centrosomes to recoil from the ablation site; mudf01205 significantly reduces centrosome recoil upon microtubule ablation (mean centrosome velocity relative to microtubule ablation site). Tricellular junctions (TCJ) specify spindle orientation in epithelial tissues via mud. Cell division orientation along the interphase cell long axis and TCJ distribution is strongly reduced in mud mutant tissue.

In mudf01205/mudf01205 pupal sensory mother cells, mitotic spindle orientation is parallel to the plane of the epithelium; the orientation of mitotic spindle is also randomized relative to the anterior-posterior axis. In around a quarter of mudf01205/mudf01205 pupal sensory mother cells in telophase, pon fails to exclusively segregate in one of the two daughter cells (pIIb to pIIa cell fate mis-specification): wild type sensory organs are composed of four cells (external socket and shaft, internal neuron and sheath) whereas mudf01205 mutant organs (in pupae 24hr APF) are composed only of external cells.

Mitotic spindle is aligned with the pins crescent is most dsh1/dsh1 pupal sensory mother cells in metaphase, but spindle fails to align in dsh1;mud4 or dsh1;mudf01205 double mutants.

Comments

Expression of mudT:Avic\GFP (but not mudΔCC.T:Avic\GFP) rescues the reduction in centrosome recoil velocity following microtubule ablation seen in mudf01205.
