Expression of duphsp70.T:Hsap\MYC results in mitotic and endocycling follicle cells with large nuclei, and an increase in the cell DNA content. Increased apoptosis is seen in mitotic follicle cells at stage 5, but not in the endocycling follicle cells of later stage egg chambers.
Expression of duphsp70.T:Hsap\MYC induces apoptosis in the germline nurse cells during their mitotic divisions in the germarium, but not after they had entered the endocycle in oogenesis stage 1.
Expression of duphsp70.T:Hsap\MYC in early third instar larvae induces apoptosis in all imaginal discs. There is no evidence of any apoptosis in any endocycling cells, such as the salivary glands, fat body and gut.
Many ovarian follicle cells in stage 5-6 egg chambers from duphsp70.T:Hsap\MYC adult females subjected to 30 minute heat shocks twice a day have enlarged nuclei. The penetrance of this phenotype is variable. A similar phenotype is seen in the imaginal discs of duphsp70.T:Hsap\MYC larvae following heat shock. 30 hours after heat shock about 50% of wing discs have some greatly enlarged nuclei. Some of thee even have small polytene chromosomes. FACS analysis of dissociated cells from these discs reveals significant numbers of cell with a a ploidy of 8C. By 54 hours after heat shock all discs have extremely aberrant morphology, some cells with enlarged nuclei, an increased level of cell death. Third instar eye discs from duphsp70.T:Hsap\MYC animals given 3 heat shock pulses over 30 hours have a significantly wider stripe of BrdU incorporating (S-phase) cells just posterior to the morphogenetic furrow than wild-type. This increase in BrDu incorporation does not extend into the differentiating cells further posterior to the furrow. These discs also have a greater number of the asynchronous cycling cells anterior to the furrow, and a low level of BrdU incorporating cells within the furrow itself. BrdU incorporation into amplification foci in stage 10B oocyte follicle cells is greatly increased in duphsp70.T:Hsap\MYC animals three hours after heat shock.
