Description

UASt regulatory sequences drive expression of a mutated form of Cdc42 which carries the V12 constitutive activation mutation and in addition, a Y40C mutation in the effector loop. The protein is tagged with two copies of Tag:MYC.

In Cdc42V12.C40.Scer\UAS.T:Hsap\MYC; Scer\GAL4elav.PLu embryos, segmental nerve (SN)a and SNb motor neuron axons (abdominal posterior fascicle) and intersegmental nerve (ISN) motor neuron axons (anterior fascicle) all exhibit normal guidance.

Expression of Cdc42V12.C40.Scer\UAS.T:Hsap\MYC in SNb neurons under the control of Scer\GAL4elav-C155 results in a normal pathfinding pattern in the majority of cases, or stalling at or before muscle 6.

SNd, SNa, and SNc axons expressing Scer\GAL4elav-C155-driven Cdc42V12.C40.Scer\UAS.T:Hsap\MYC display wild-type pathfinding patterns in 100% of cases.

Expression of Cdc42V12.C40.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4elav.PLu increases LBD filopodial activity, with extension increased by 32% and retraction by 43%, respectively.

SNb growth cones expressing Cdc42V12.C40.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4elav.PLurevert to wild-type pathfinding, but show no significant change in filopodial activity from wild-type. The only exception is a 20% increase in the number of filopodia directed posteriorly.
