Gitex10/Gitf03586 flies have significant defects in climbing ability compared to controls. Postsynaptic glutamate receptor fields are enlarged in neuromuscular junctions of Gitex10/Gitf03586 third instar larvae. Gitex10/Gitf03586 third instar larval NMJs (type 1b) are similar to controls in terms of synaptic vesicle number, docked vesicle number, active zone size or T-bar morphology; however, stimulated mutants display an accumulation of endosomal structures and vacuoles (>80nm vesicles or cisternae). Gitex10/Gitf03586 third instar larval NMJs show significant reductions in evoked excitatory junctional currents (eEJCs) compared to controls (no difference in rise times or decay constants). Gitex10/Gitf03586 larval NMJs show defects in synaptic vesicle recycling, and specific defects in the slow phase of endocytosis and synaptic vesicle reacidification. Under conditions of elevated extracellular calcium, evoked neurotransmission of Gitex10/Gitf03586 larvae is similar to controls but FM1-43 uptake is significantly reduced (impaired endocytosis or synaptic vesicle recycling).
