At 20oC, expression of brkScer\UAS.cMa in the posterior of the wing disc, driven by Scer\GAL4en-e16E, produces a mild phenotype in adult wings. The posterior of the wing is slightly reduced compared to wild type and wing veins 4 and 5 are incomplete at the distal end. When flies of the same genotype are raised at 25oC, the wing phenotype is more severe; the posterior is more reduced in size, the wing margin is notched distally, vein 4 and the posterior crossvein are lost, and vein 5 is incomplete distally. Examination of wing discs of this genotype shows that disc shape is altered relative to wild type; at 25oC, the discs have a deep invagination between brkScer\UAS.cMa-expressing cells in the posterior and non brkScer\UAS.cMa-expressing cells in the anterior. At 20oC, this effect is reduced, but a small invagination is still observed.

The wing defects seen in flies expressing brkScer\UAS.cMa are not due to excessive cell death as preventing cell death by overexpression of BacA\p35Scer\UAS.cHa does not suppress the phenotype. Flies co-expressing brkScer\UAS.cMa and BacA\p35Scer\UAS.cHa, both under the control of Scer\GAL4en-e16E, at 20oC show the same phenotype as flies that express only brkScer\UAS.cMa. When brkScer\UAS.cMa and BacA\p35Scer\UAS.cHa are co-expressed at 250C, again driven by Scer\GAL4en-e16E, there is a slight blistering of the adult wing at the posterior, indicating that extra wing tissue is present when cell death is prevented. However, the wing vein defects and notching still occur.
