Description

UAS regulatory sequences drive expression of a CaMKII hairpin. The hairpin corresponds to a 612bp CaMKII cDNA sequence ligated to a 528bp CaMKII inverted sequence that matches the longer fragment from the 5' end onwards.

Expression of CaMKIIdsRNA.Scer\UAS under the control of Scer\GAL4VGlut-OK371 results in a decrease in bouton number/area for type Ib boutons at the larval neuromuscular junction.

Flies expressing CaMKIIdsRNA.Scer\UAS under the control of either Scer\GAL4ey-OK107, Scer\GAL4c305a, Scer\GAL4Mef2.247 or Scer\GAL4CaMKII.7 show normal learning (measured as 2 minute memory) in an olfactory aversive conditioning assay.

Flies expressing CaMKIIdsRNA.Scer\UAS under the control of either Scer\GAL4ey-OK107, Scer\GAL4c305a or Scer\GAL4Mef2.247 show normal middle-term memory (measured 3 hours after one cycle of training) in an olfactory aversive conditioning assay.

Flies expressing CaMKIIdsRNA.Scer\UAS under the control of either Scer\GAL4ey-OK107, Scer\GAL4c305a or Scer\GAL4Mef2.247 show severely reduced long-term memory (measured 24 hours after 5 cycles of spaced training) in an olfactory aversive conditioning assay.

1 day memory is impaired after spaced training, but not after massed training in an olfactory associative conditioning assay in flies expressing CaMKIIdsRNA.Scer\UAS under the control of Scer\GAL4G0431. The defect in memory after spaced training is still seen if the temperature sensitive Scer\GAL80ts.αTub84B allele is used to restrict Scer\GAL4 expression to the adult stage.

Flies expressing CaMKIIdsRNA.Scer\UAS under the control of Scer\GAL4CaMKII.7 show a near-complete loss of long-term olfactory memory, while short-term memory is unaffected.
