Description

An inverted repeat corresponding to nucleotides 634-1033 of the dco coding sequence, separated by an intron from the w gene, is under the control of UAS sequences. The dco sequences included in the construct are not predicted to interfere with any of the other eight CkI homologs.

Expression of dco^{dsRNA.WIZ.Scer\UAS}, under the control of Scer\GAL4^ptc-559.1, at 29^oC results in a narrowing between wing veins 3 and 4. Scer\GAL4^ptc-559.1-driven expression at 25^oC results in the mispolarization of wing hairs between veins 3 and 4. Polarity swirls can be observed extending 4% of the distance between the posterior cross vein and the distal tip of the wing.

Coexpression of CkIα^{dsRNA.WIZ.Scer\UAS} enhances the Scer\GAL4^ptc-559.1>dco^{dsRNA.WIZ.Scer\UAS} wing phenotype at 25^oC, so that the wing hair polarity swirls extend to 17% of the wing length from the posterior cross vein, rather than the 4% seen with solo expression of dco^{dsRNA.WIZ.Scer\UAS}.
