UASt sequences drive expression of a genomic fragment encoding amino acids 11--337 of the CkIα gene with the corresponding cDNA fragment inserted in a reverse orientation. The dsRNA produced knocks down expression of CkIα and also of dco (the dsRNA fragment includes a 30bp contiguous sequence that is shared between CkIα and dco).
Transcription from the P{UAS-CkIα.CRL.dsRNA} construct should produce CkIα dsRNA, resulting in dsRNA interference (RNAi) of the CkIα gene.
The P{UAS-CkIα.CRL.dsRNA} construct provides a more efficient knockdown of CkIα than the P{UAS-CkIα.RNAi} construct.
This construct results in an effective knockdown of the CkIα gene, but also results in a partial knockdown of the dco gene.
