Description

UASt regulatory sequences drive expression of an active form of SPE which does not require post-translational processing. This is achieved by fusing a signal peptide (Tag:SS(ea)) to the SPE catalytic domain (amino acids 135-400). The protein is also tagged with Tag:V5.

Misexpression of SPEactive.Scer\UAS.T:SV5\V5 either in the fat body (Scer\GAL4Lsp2.PH), or hematopoietic (Scer\GAL4Hml.PG) or both (Scer\GAL4Cg.PA) induces cell aggregation and melanization upon L.victoriae parasitic wasp infection.

Expression of SPEactive.Scer\UAS.T:SV5\V5 under the control of Scer\GAL476B significantly increases the infiltration index of blood cells into the fat body, indicating an increase in inflammatory immune response.
