UASt regulatory sequences drive expression of an inverted repeat.
Expression of FitmGD3580 under the control of either Scer\GAL4αTub84B.PL or Scer\GAL4Mef2.PR leads to reduced climbing capability and flightlessness in both young and older flies, expression under Scer\GAL4C7 impairs in older flies only (both climbing and flight ability is comparable to controls at 4 days after eclosion). Expression driven by either Scer\GAL4elav.PLu or Scer\GAL4nSyb.PU (both with UAS-Dicer2 to enhance RNAi efficiency) does not have significant effect on the adult climbing or flight ability regardless of age.
A subset of larvae expressing FitmGD3580 (together with UAS-Dicer2) under the combined control of Scer\GAL4ppk.PG and Scer\GAL4477 display a strong reduction in the dendritic field coverage (lower maximum radius, reduced average as well as cumulative branch path and decreased branch number) in dorsal multidendritic ddaC neurons compared to controls.
Scer\GAL4αTub84B.PL or Scer\GAL4elav.PLu-driven expression of FitmGD3580 impairs adult hearing: the threshold for sound-evoked compound action potential is increased, the mechanical amplification gain (provided by auditory sensory neuron motility) is reduced and the best frequency of the antennal sound receiver is higher relative to controls. Expression under the Scer\GAL4C7 driver results in significantly diminished size of lipid droplets in the fat body of both young and older adult flies.
Adults expressing CG10671GD3580 under the control of the cardioblast-specific Scer\GAL4tin.CΔ4 driver show significantly reduced survival on day 6 after a shift to 29[o]C compared to control flies.
Adults expressing CG10671GD3580 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
