CnbGD11735 driven by Scer\GAL4wor.PU does not affect brain volume, neuroblast number, or cell cycle length in third instar larvae.
Immediately after mitosis, centrosome migration to the apical cortex and organisation of a prominent interphase microtubule organising center (MTOC) occur in larval neuroblasts expressing CnbGD11735 under the control of Scer\GAL4Act5C.PI, as occurs in wild type. However, during interphase both centrioles become motile and no major MTOC is observed in the mutant cells (in contrast to wild type, where one centriole retains MTOC activity and remains apical-cortex bound and the other is motile). Asymmetric mitosis is accomplished in the mutant neuroblasts, but the site of ganglion mother cell budding is not maintained from one cycle to the next as efficiently as it is in controls.
Adults expressing CnbGD11735 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
