UASt regulatory sequences drive expression of an inverted repeat.
Expression of Slc25A46GD1757 under the control of Scer\GAL4elav.PLu results in reduced locomotor ability in both third instar larvae (decreased crawling velocity) and adults (reduced climbing ability) but the adult viability is not significantly affected. The knock-down larvae also display morphological defects at the neuromuscular junction (NMJ): decreased total branch length as well as reduced number and size of boutons; the density and size of presynaptic active zones (visualized by brp staining) appear to be decreased too but the difference is not statistically significant. The size of mitochondria at the NMJ in the Scer\GAL4Toll-6-D42>Slc25A46GD1757 larvae on the other hand is increased but their total number is comparable to controls.
Larvae expressing Slc25A46GD1757 under Scer\GAL4elav.PLu also display aberrations in the targeting of R7-R8 photoreceptor axons into medulla, the diameter/length ratio of the optic stalk is not significantly affected.
Adults expressing CG5755GD1757 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
