UASt regulatory sequences drive expression of an inverted repeat.
Expression of Msp300GD10317 under the control of Scer\GAL4how-24B (muscle-specific) but not Scer\GAL4elav.PU (neural) leads to larval locomotion deficit (reduced crawling ability - distance crawled/time) and also clustering of the muscle nuclei. The effect is further strengthened by addition of UAS-Dicer2 to enhance the RNAi efficiency.
Adults expressing Msp300GD10317 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
