Coexpression of Or43aScer\UAS.T:Avic\GFP-YFP(1) and Or83bScer\UAS.T:Avic\GFP-YFP(2), under the control of Scer\GAL4Gr21a.9.323, reconstitutes a functional odorant receptor as measured in neurons that also express the calcium-sensitive reporter Ggal\MLCK::Rat\CamG-CaMP.Scer\UAS.T:Avic\GFP-cpEGFP. Expression of Or43aScer\UAS.T:Avic\GFP-YFP(1) and Or83bScer\UAS.T:Avic\GFP-YFP(2) results in odor-evoked calcium responses to cyclohexanol and benzaldehyde across broad domains of the antennal lobe, while expression of Ggal\MLCK::Rat\CamG-CaMP.Scer\UAS.T:Avic\GFP-cpEGFP alone results in only sparse activation patterns.
This construct is designed for a "protein-fragment complementation assay (PCA)"; the T:Avic\GFP-YFP C-terminal fragment is complementary to an N-terminal T:Avic\GFP-YFP fragment which is carried on separate constructs. Binding of proteins tagged with either of the two T:Avic\GFP-YFP fragments brings them in close enough proximity to fold into an active form and fluoresce.
