CdsACB-0128-3 homozygous male adults as well as CdsACB-0128-3/Df(3L)BSC795 are completely sterile but CdsA1/CdsACB-0128-3 males are fertile.
Adult CdsACB-0128-3 homozygote males have no mature sperm in the seminal vesicles and display spermatid individualization defects: the actin-rich investment cones are established but are disorganized and move asynchronously along the spermatids and neither a cystic bulge nor a proper waste bag is observed.
Adult CdsACB-0128-3 homozygote males do not display any defects in the morphology of mitochondria in primary spermatocytes, early round spermatids or elongating spermatids: the mitochondrial fusion, nebenkern formation as well as elongation is normal during these stages. However, elongated pre-individualized CdsACB-0128-3 cysts contain disorganized spermatids with either same-sized mitochondrial derivatives or enlarged, different-sized derivatives and also show overgrowth of the axonemal sheath.
Adult CdsACB-0128-3 homozygote males do not display normal climbing and flight abilities compared to wild-type.
CdsCB-0128-3 has male sterile phenotype, suppressible | partially by PisβTub85D.PL
CdsCB-0128-3 has spermatid phenotype, suppressible | partially by PisβTub85D.PL
CdsCB-0128-3 has waste bag phenotype, suppressible | partially by PisβTub85D.PL
The adult male sterility characteristic for CdsACB-0128-3 homozygotes is partially rescued by expression of PisβTub85D.PL in the mutant background (the number of dispersed actin cones in the elongated spermatids is reduced and the number of waste bags is increased compared to CdsACB-0128-3 mutants).
CdsCB-0128-3 is rescued by CdsUASp.EGFP
Expression of CdsAScer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4bam.T:Hsim\VP16 rescues the spermatid individualization defects and adult male sterility of CdsACB-0128-3 homozygotes.
Precise excision of the P{RS3} element reverts the male sterile phenotype.
