UASt regulatory sequences drive expression of an inverted repeat.
viable, with Scer\GAL4G7
visible | adult stage | progressive, with Scer\GAL4da.PU
Expression of wmdKK100974 under the control of either Scer\GAL4Mef2.PR or Scer\GAL4elav.PU (both with Dcr-2Scer\UAS.cUa) or driven by any of the following: Scer\GAL4c179, Scer\GAL4how-24B or Scer\GAL4da.PU (all without Dcr-2Scer\UAS.cUa) reduces adult viability to a varying degree (up to a point of complete lethality in some cases) at both 25[o]C and 29[o]C with more severe effects being observed at the higher temperature. Expression under Scer\GAL4tub.PU is completely lethal at both temperatures while expression under either Scer\GAL4G7 or Scer\GAL4RapGAP1-OK6 (with or without Dcr-2Scer\UAS.cUa) has practically no effect on viability.
Adult escapers expressing wmdKK100974 under the control of Scer\GAL4da.PU show wing defects.
Adult flies expressing wmdKK100974 (together with Dcr-2Scer\UAS.cUa to enhance RNAi efficiency) under the control of Scer\GAL4elav.PU are short lived and show age-progressive loss of flight ability compared to controls. This decline in flight performance is not observed when the Scer\GAL4Mef2.PR driver is used (without Dcr-2Scer\UAS.cUa).
No decline in adult flight ability compared to controls is observed when wmdKK100974 and Gem3ΔN.Scer\UAS are co-expressed under the Scer\GAL4Mef2.PR driver.
