Deletion of most of the protein coding sequence of Su(var)3-7 (81 amino acids from the N-terminus remain).
The progeny of homozygous mutant flies stop developing during the second larval instar. This maternal effect is rescued by a paternal wild-type copy of Su(var)3-7, but only half of the expected male progeny are recovered. Zygotically mutant male and female animals show approximately 90% viability.
Mutant larvae show an extreme enlargement of the ring gland due to an increase in size of the prothoracic gland cells. The polytene chromosomes of the prothoracic gland cells are more highly endoreplicated than normal.
The sumoylation of Su(var)3-7 (at the lysine at position K839) is necessary for heterochromatic localization and the ability to enhance position-effect variegation.
