Gene Group (FlyBase)
MADF-BESS DOMAIN TRANSCRIPTION REGULATORS -
The MADF-BESS domain transcription regulators include a variety of activators and corepressors and chromatin modifying proteins. BESS domains are often present in combination with MADF domains. The BESS domain mediates protein-protein interactions. (Adapted from FBrf0155780 and FBrf0224045).
Protein Function (UniProtKB)
Dose-limiting factor in position-effect variegation, the inactivation in some cells of a gene translocated next to heterochromatin. It could play a role in chromosome condensation.
(UniProt, P20193)
Summary (Interactive Fly)

chromatin constituent - C2H2 zinc finger protein - required for X-restricted dosage compensation complex targeting - increasing the amount of Su(var)3-7 triggers heterochromatin expansion and epigenetic gene silencing.

Structure
Protein 3D structure   (Predicted by AlphaFold)   (AlphaFold entry P20193)

If you don't see a structure in the viewer, refresh your browser.
Model Confidence:
  • Very high (pLDDT > 90)
  • Confident (90 > pLDDT > 70)
  • Low (70 > pLDDT > 50)
  • Very low (pLDDT < 50)

AlphaFold produces a per-residue confidence score (pLDDT) between 0 and 100. Some regions with low pLDDT may be unstructured in isolation.

DNA-protein interactions: genome-wide binding profile assayed for Su(var)3-7 protein in Kc167 cells; see Chromatin_types_NKI collection report. Individual protein-binding experiments listed under "Samples" at GEO_GSE22069 (http://www.ncbi.nlm.nih.gov/geo/query/acc.cgi?acc=GSE22069).

The genomic organisation and pattern of expression of the Su(var)3-7 locus have been characterised.

Phenotype of wbl is unchanged by Su(var)3-7.

Su(var)3-7 is a haplo suppressor and a triplo enhancer. The suppressor effect of Su(var)3-9 cannot dominate the enhancer effect displayed by two extra doses of Su(var)3-7.

Deletion of one wild type copy of Su(var)3-7 reduces In(1)wm4 position effect variegation and duplication of Su(var)3-7 enhances variegation. A 6.5kb EcoRV gene fragment from Su(var)3-7 is capable of enhancing variegation in a dose-dependent manner.

Duplications of Su(var)3-7 were found to considerably reduce the haplo-abnormal effect of heterozygous Su(var)3-7 point mutations.

        Additional comments

Complements all lethals in the region except Acej41; Acej41 has a weak dominant variegation suppression effect.
