An EP-element insertion that disrupts the 5' UTR of both predicted transcripts of the RpS5b gene.
RpS5bG5346 homozygous females are sterile and do not complete oogenesis. Ovaries are small. Nurse cells, follicle cells, and oocyte differentiate, but very few egg chambers progress beyond stage 8-9, after which point apoptosis is induced (cleaved caspase-3). Some egg chambers contain more than 16 germ cells with one or two oocytes, some egg chambers fail to separate, while others have 16 germ cells but two oocytes, or a mis-localized oocyte. There are increased numbers of follicle cells, with multi-layering at the posterior of egg chambers. Stage 7 oocytes show polarity defects, as microtubules are improperly aligned and often accumulating by the anterior of the oocyte, instead of at the posterior, and Oskar and Gurken are not properly localized; oocytes also show increased F-actin levels.
RpS5bG5346 germline clones also show severe oogenesis defects. Many egg chambers have multiple layers of follicle cells, and encapsulation defects are observed.
Ovaries from RpS5bG5346 homozygotes or germline clones show extensive mitochondria clustering, in contrast to a more dispersed distribution in controls. In most cases, mitochondria are less elongated and have less well resolved cristae; sometimes two mitochondria adhere to each other via the outer membrane, and some mitochondria bend and engulf some cytoplasm, forming a donut shape.

Expressing RpS5aGL01502 under the control of Scer\GAL4VP16.nos.UTR in a RpS5bG5346 homozygous background leads to a much more severe oogenesis phenotype than in RpS5bG5346 homozygotes alone: ovaries are only very rudimentary and no formation of egg chambers is apparent.
