Imprecise excision of the progenitor insertion, resulting in a 1.4kb deletion that removes the first exon and part of the second exon of RASSF8. 81bp of the 5' untranslated region of the neighbouring tankyrase gene is also deleted, but the level of the tankyrase transcript is not obviously affected.
RASSF86 mutants have slightly rounder wings and show a significant increase in the number of inter-ommatidial cells in the pupal eye disc.
Homozygotes have a broad wing phenotype, with the wings being shorter along the proximodistal axis and broader along the anteroposterior axis compared to wild-type controls.
The average wing area of homozygotes is 5.8% larger than that of controls, while the wing hair density is comparable to that of controls, indicating that the wings contain more cells rather than larger cells. The adult body weight of homozygous flies is 10.5% higher than that of control flies.
Homozygotes have enlarged, rough eyes. The hexagonal ommatidial lattice is disrupted and there is clustering of interommatidial bristles. Extra interommatidial cells are seen in the retinas of mutant pupae, with a double layer of these cells often being seen. The mutant pupal retinas show a significant decrease in apoptosis compared to controls. Homozygous clones in the pupal retina also show a significant decrease in apoptosis compared to control clones.
RASSF8[+]/RASSF86 is a suppressor of visible | adult stage phenotype of MagiUAS.GFP, Scer\GAL4GMR.PU
RASSF8[+]/RASSF86 is a suppressor of eye | adult stage phenotype of MagiUAS.GFP, Scer\GAL4GMR.PU
The rough eye phenotype in adult flies induced by expression of MagiScer\UAS.T:Avic\GFP under the control of Scer\GAL4GMR.PU is significantly suppressed by combination with a single copy of RASSF86.
RASSF86 is rescued by RASSF8Ubi-p63E.PZ
RASSF86 is rescued by RASSF8UASp.cLa/Scer\GAL4arm.PS
RASSF86 is partially rescued by RASSF8YA.Ubi-p63E
The increased number of inter-ommatidial cells characteristic for RASSF86 flies is fully rescues by expression of RASSF8Ubi-p63E.PZ in the mutant background.
