5.0kb HindIII-HindIII genomic fragment which includes the entire RacGAP84C transcription unit together with approximately 1.5kb each of upstream and downstream genomic sequence.
RacGAP84Ct5.0 partially rescues Df(3R)rn20
Expression of RacGAP84Ct5.0 partially rescues the defects seen in the testes of homozygous Df(3R)rn20 males; the overall length of the cysts is significantly increased and the "ball-of-yarn" phenotype seen at the end of the cysts in Df(3R)rn20 males is significantly reduced. Orientation of spermatids is restored almost to normal in the rescued males, although cysts remain somewhat larger than normal and fail to attain paracrystalline packing. The morphology of individual spermatids is almost completely rescued to normal. The rescued sperm are motile, as the seminal vesicles of the rescued males contain motile sperm, in contrast to those of Df(3R)rn20 homozygotes. However, the fertility of the rescued males is only 5-10%.
The retinal defects seen in Df(3R)rn20 homozygotes are rescued by RacGAP84Ct5.0; the ommatidia are well organised and contain the normal number of full-length photoreceptor cells surrounded by pigment cells. The pigment cell layer appears sparser than that of wild type.
