UAS regulatory sequences drive expression of a genomic fragment consisting of part of exon 2 (the last 654bp), exon 3, part of exon 4 (the first 18bp) and introns 2 and 3 of Tk fused to the corresponding inverted cDNA fragment.

Male and female flies expressing TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4c739 cross the centre of the assay arena less often than controls in an assay of walking behaviour. The total distance moved, number of start/stop actions and mean walking speed are not significantly different from wild type.

Female flies expressing TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4121Y cross the centre of the assay arena less often than controls in an assay of walking behaviour.

Female but not male flies expressing TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4NP2320 display an increase in the number of start/stop actions compared to controls during a walking assay.

Expression of TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4c205 has no significant affect on locomotor behaviour in a walking assay.

Expressing TkdsRNA.UAS.cWa under the control of either Scer\GAL4dimm-929 or Scer\GAL4Kurs6 results in a reduction in lifespan when flies are deprived of food alone or both food and water. When deprived of food and water, flies expressing TkdsRNA.UAS.cWa under the control of Scer\GAL4Kurs6 lose significantly more water than controls.

Flies expressing TkdsRNA.UAS.cWa under the control of Scer\GAL4Kurs6 have normal locomotor activity even when starved.

Expression of TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4αTub84B.PL results in lethality; 97% of the animals die at the embryonic stage, and 3% die as first instar larvae.

Adults expressing TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4elav-C155 and controls flies show a similar response to acetone across a range of concentrations tested in an olfactory choice assay. However, animals expressing TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4elav-C155 are less repelled than control flies by benzaldehyde or butanol at high concentrations. Animals expressing TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4elav-C155 show significant differences in response to isoamyl acetate compared to control flies over a wider range of concentrations.

Larvae expressing TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4elav-C155 do not show attraction to benzaldehyde or isoamyl acetate in an olfactory choice assay, in contrast to controls.

Adults expressing TkdsRNA.Scer\UAS.cWa under the control of Scer\GAL4elav-C155 show higher locomotion activity than control flies in the Buridan's paradigm (a locomotion test based on the walking activity between two opposing visual stripes). The percentage of active time and distance walked are significantly increased, while the speed of movement is not altered compared to controls.
