Description

UASp regulatory sequences drive expression of a cnn coding sequence which lacks the amino terminal 17 amino acids of the maternal cnn-RA transcript and in which 13 core amino acid residues in the conserved "motif 1" have been deleted. The cnn sequences are tagged with EGFP.

Embryos expressing cnnΔ1.Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 do not hatch. Centrosome duplication appears normal in these embryos, but centrosome separation fails. The mutant embryos also have reduced numbers of centrosomal satellites.

Expression of cnnΔ1.Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4tub.PU from late embryogenesis onwards is not zygotically lethal.
