Description

Contains a 4.5kb XhoI fragment containing the lid upstream region and part of the coding region and a 4.8Kb XhoI/NotI cDNA fragment containing the remainder of the lid coding sequence, which has been mutated to contain two point mutations in the JmjC domain that prevent Fe[2+] binding (amino acid replacements H637A and E639A).

lidunspecified mutant males rescued by lidgJmjC.mut have a significantly shorter lifespan that wild-type controls, while lidunspecified mutant females rescued by lidgJmjC.mut have a mean lifespan that is not significantly different to wild type.

lidunspecified mutant males rescued by lidgJmjC.mut are sensitive to paraquat compared to controls, while lidunspecified mutant females rescued by lidgJmjC.mut have normal sensitivity to paraquat.

Expression of lidgJmjC.mut rescues the karyosome, centromere clustering and synaptonemal complex defects seen in the ovaries of lidk06801/lid10424 mutants.

Expression of lidgJmjC.mut partially rescues the locomotor rhythm defects seen in homozygous lid10424 mutant flies. Under LD (12 hours of light:12 hours of dark) and DD conditions (constant darkness) flies exhibit attenuated day:night differences in activity, with flies showing increased activity around midday and reduced dark anticipation.
