Expression of ana2Ubi.C.GFP in combination with Sas-6Ubi.P.GFP but not with Poc1UAS.B.GFP (under the control of Scer\GAL4VP16.bam) leads to the formation of the procentriolar ectopic particles that are located in the cytoplasm of mature and immature primary spermatocytes, and attached to the centrioles only in mature primary spermatocytes when compared to controls. Procentriolar ectopic particles in mature primary spermatocytes resemble the proximal centriole-like ultrastructure with tubule-like structures, spokes, and electron-dense material.
Expression of ana2Ubi.C.GFP in combination with Sas-6Ubi.P.GFP leads to formation of procentriolar ectopic particles while their expression in Poc1W87X homozygous background leads to structural defects in procentriolar ectopic particles such as fragmented tubule-like structures and electron-dense material in mature primary spermatocytes.
Almost all unfertilized eggs laid by mothers carrying one copy of sas-6Ubi.P.T:Avic\GFP and ana2Ubi.C.T:Avic\GFP contain hundreds of large structures that stain for centriole markers, recruited PCM, and nucleated asters. These centriole-like structures often appear ring-shaped, and only one structure is contained within each aster. Eggs laid by mothers carrying one copy of either ana2Ubi.C.T:Avic\GFP or sas-6Ubi.P.T:Avic\GFP together with one copy of either SAKUbi.P.T:Avic\GFP or Sas-4Ubi.P.T:Avic\GFP exhibit at most a very small number of asters in very few eggs.
