An upstream 4209bp Hira fragment (consisting of Hira upstream sequences) and a downstream 3530bp Hira fragment (containing genomic sequence from the second exon through to the end of the transcription unit) are cloned either side of the w^+mW.hs marker gene which is flanked with stop codons in all 6 reading frames. Three copies of the Tag:FLAG tag have been inserted at the C-terminal end of the Hira coding sequence in the downstream arm. Transcription occurs from the Hsp70 promoter present in the w^+mW.hs marker gene, resulting in a transcript that contains Hira sequences from the downstream fragment, but no Hira-Tag:FLAG protein is detected.
