UAS regulatory sequences drive expression of a short inverted repeat.
Scer\GAL4btl.PU-mediated expression of expHMS01445 produces no detectable defects in the pattern of migration, organisation, or diversification of tracheal cells. However, a clear defect in chitin deposition is detected - luminal chitin accumulation is prevented in dorsal and ventral branches, remaining only in the dorsal trunk and part of the transverse connective. Only the branches that accumulate chitin fill with gas.
Scer\GAL4tub.PU- or Scer\GAL469B-mediated expression of expHMS01445 results in inflated and shorter embryos that show defects in the head skeleton. 20% of embryos show defective mouth inversion or rotation.
Scer\GAL4btl.PU, expHMS01445 has embryonic/larval tracheal section phenotype, non-suppressible by Scer\GAL4btl.PU/kkvUAS.GFP
Scer\GAL4btl.PU-mediated expression of expHMS01445 in a Df(2R)BSC329/Df(2R)BSC879 background (that removes reb) results in embryos with a normal branching pattern but devoid of the chitin filament. Branches do not fill with air a the end of embryogenesis and embryos die.
