Description

The per CRS (circadian regulatory sequence) and a basal Hsp70 promoter are fused upstream of sequences encoding the per coding region (a hybrid of cDNA and genomic sequences). The genomic portion includes the 3' UTR, including the 3' terminal intron. XhoI and KpnI sites have been introduced flanking this 3' terminal intron.

      Transgenic per8:8 flies in a per01 mutant genetic background exhibit sleep patterns very similar to those previously described for wild-type strains, including the temperature and gender differences on daytime sleep.

      per01 flies rescued with either perDyak\per.3' or perM2M1 generally show earlier onsets of evening activity and shorter, less robust, midday siestas compared to per01 flies rescued with per8:8.
