Df(3R)d189 homozygotes or Df(3R)d189/Df(3R)P14 animals in which the function of all genes affected by Df(3R)d189 except Ada2a have been rescued using P{CaSpeR4-Dtl+Rpb4+} are either late-larval of early-pupal lethals. The animals follow a seemingly normal course of development until the third larval instar stage, but approximately a quarter of them (27%) never pupate. Three quarters (73%) form prepupae, which are similar to larvae in shape, are covered with a brownish cuticle, and die at this stage.

Females with Df(3R)d189/Df(3R)P14 P{CaSpeR4-Dtl+Rpb4+} germlines are not recovered in experiments transplanting mutant germline cells into host embryos, indicating that Ada2a function is required in the germline.

In disc-derived body-parts, homozygous Ada2ad189 clones (homozygous for Df(3R)d189 with the function of all genes affected by the deficiency except Ada2a having been rescued using P{CaSpeR4-Dtl+Rpb4+}) develop as frequently as control clones, but are significantly smaller than control clones. Homozygous clones on the abdomen are as large as control clones.
