UASt regulatory sequences drive expression of the Tag:SS(nec) secretory signal peptide followed by two tandem copies of the Aβ (amyloid β) monomer. Both copies of Aβ are the 40 amino acid residue form. The two copies are directly fused, with no linker sequence between them.
Flies expressing Hsap\APPTNL.Aβ40.Scer\UAS under the control of Scer\GAL4GMR.PF form significant quantities of large, SDS-insoluble deposits in the head. Very few SDS-soluble oligomeric Aβ aggregates are present.
Expression of Hsap\APPTNL.Aβ40.Scer\UAS under the control of Scer\GAL4GMR.PF does not result in a detectable rough eye phenotype.
Flies expressing one copy of Hsap\APPTNL.Aβ40.Scer\UAS under the control of Scer\GAL4elav-C155 do not have a significantly different life span compared to wild-type controls.
