UASt regulatory sequences drive expression of the Tag:SS(nec) secretory signal peptide followed by two tandem copies of the Aβ (amyloid β) monomer linked together by a 22 amino acid linker peptide. Both copies of Aβ are the 40 amino acid residue form.
Flies expressing Hsap\APPT22.Aβ40.Scer\UAS under the control of Scer\GAL4GMR.PF form significant quantities of large, SDS-insoluble deposits in the head. Abundant SDS-soluble oligomeric Aβ aggregates are also present. No detectable SDS-soluble oligomeric Aβ aggregates are present.
Expression of Hsap\APPT22.Aβ40.Scer\UAS under the control of Scer\GAL4GMR.PF does not result in a detectable rough eye phenotype.
Flies expressing one copy of Hsap\APPT22.Aβ40.Scer\UAS under the control of Scer\GAL4elav-C155 do not have a significantly different life span compared to wild-type controls.
