Mpc11/Mpc12 mutants are sensitive to a carbohydrate-only diet, dying shortly after transfer to a sucrose medium. The amount of adenosine 5'-triphosphate (ATP) is reduced, along with triacylglycerol (TAG) and protein. The amounts of carbohydrates are elevated, including the circulating sugars trehalose, glucose, fructose and glycogen. Pyruvate levels are elevated, whereas levels of TCA cycle intermediates (citrate, isocitrate, succinate, fumerate and malate) are reduced compared to controls. Increased levels of glycine and serine are seen and glutamate, aspartate and proline are all depleted.

Expression of Mpc1Scer\UAS.cBa under the control of Scer\GAL4tub.PU rescues the lethality seen in Mpc11/Mpc12 mutant flies on a sucrose diet. The lethality is also rescued when Mpc1Scer\UAS.cBa is expressed in the fat body (Scer\GAL4Cg.PA), muscle (Scer\GAL4Mef2.PR) or neurons (Scer\GAL4elav.PLu). No rescue is seen when Mpc1Scer\UAS.cBa is expressed in the gut under the control of Scer\GAL4mex1.2.1.
