Detailed Description

fonΔ17A homozygous and fonΔ17A/Df(2L)Exel6043 transheterozygous pupae are elongated, as shown by a significant increase in the length/width axial ratio as compared to controls.

fonΔ17A/Df(2L)Exel6043 transheterozygous wandering third instar larvae present significant detachment of myofibers and a significant increase in the hemisegmental gap between dorsal oblique muscles 1 and 2, as compared to controls; type I synapses enervating lateral longitudinal muscle 1 present similar bouton number, as compared to controls.

At the muscle attachment sites of fonΔ17A/Df(2L)Exel6043 transheterozygous wandering third instar larvae, muscle and tendons are adjacent but not interdigitated and there is decreased amount of extracellular matrix in-between, tendon cells display a decrease in junctional complexes, and muscle attachment fibers present a convoluted arrangement, as compared to controls.

Hemolymph from homozygous wandering larvae produces longer fibres than can be drawn from wild-type larvae in a drawout assay testing coagulation.

Comments

Expression of fonScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4da.G32 partially rescues the lethality of fonΔ17A homozygotes. However, the increased strand length that can be drawn out from the larval hemolymph in a drawout assay testing coagulation is not rescued in these animals.
