Description

UASp regulatory sequences drive expression of a mutated form of E2f1 which contains the amino acid replacements I153A, Y156A and Y157A. Four amino acids in the RRXYD motif have also been replaced by alanine (R292A, R293A, Y295A, D296A). The protein is also tagged with EGFP.

The rate of proliferation in trypsin-dissociated posterior wing disc cells expressing E2fPIP-3A-DBD.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4en-e16E is similar to controls. A comparable proportion of cells have a G1 DNA content compared to controls.

Expression of E2fPIP-3A-DBD.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4en-e16E induces apoptosis in the posterior compartment of intact third instar larval wing discs. A similar increase in apoptosis is seen when the wing disc cells are trypsin-dissociated and quantified with flow cytometry.

The cells of the second mitotic wave in flies expressing E2fPIP-3A-DBD.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4GMR.PU show the stereotypical pattern of S phase seen in controls. As in wild type very few cells display DNA damage, however an increase in apoptosis is seen in the cells posterior to the morphogenetic furrow.
