Description

Excision of the P{lacW} element in P{lacW}Fick11101 has resulted in deletion of the 5'UTR and first 260 amino acids of Fic, including the highly conserved tetratricopeptide repeat domain.

Homozygous Fic55 mutant flies are not attracted to the light source in phototaxis assays. This defect could not be rescued by feeding flies a diet enriched with either histamine or carcinine.

Electroretinograms show that Fic55 mutant flies undergo normal light-induced depolarisation. However, they lack the ON transient, which reflects synaptic activation of the postsynaptic laminar neurons. Partial restoration of ON transients is achieved through feeding flies a diet enriched with either histamine or carcinine.

Electroretinograms show that Fic55/Df(2L)BSC296 mutant flies undergo normal light-induced depolarisation. However, they lack the ON transient, which reflects synaptic activation of the postsynaptic laminar neurons.

No defects in the arrangement of photoreceptor axons or rhabdomeres are evident in Fic55 mutants. Mitochondria, synaptic vesicles and active zone T bars also appear unchanged.

Flies containing Fic55 mutant whole-eye clones (generated using the eyFLP method) exhibit normal ON transients as seen in controls. Response to light in phototaxis assays also appears normal.

Fic55 mutant glial cells have a slightly reduced frequency of capitate projections compared to controls, with ~2.8 projections per synaptic cross section compared to approximately 4.3 in wild type. The shape and average diameter of the projections are similar to controls.

Expression of Fic+tRa rescues the phototaxis defects seen in homozygous Fic55 mutant flies. The loss of the ON transients in electroretinograms is also rescued.

Expression of FicScer\UAS.cRa under the control of Scer\GAL4arm.PS rescues the loss of the ON transients in Fic55 mutant electroretinograms.

Expression of FicScer\UAS.T:Avic\GFP under the control of Scer\GAL4arm.PS rescues the loss of the ON transients in Fic55 mutant electroretinograms.

Expression of FicScer\UAS.T:Arus\HRP under the control of Scer\GAL4arm.PS rescues the loss of the ON transients in Fic55 mutant electroretinograms.

Expression of FicH375A.Scer\UAS under the control of Scer\GAL4arm.PS fails to rescue the loss of the ON transients in Fic55 mutant electroretinograms.

Expression of FicScer\UAS.cRa under the control of Scer\GAL4elav-C155 fails to rescue the loss of the ON transients in Fic55 mutant electroretinograms. The defects in phototaxis seen in Fic55 mutants are also not rescued.

Expression of FicScer\UAS.cRa under the control of Scer\GAL4repo rescues the loss of the ON transients in Fic55 mutant electroretinograms. The defects in phototaxis seen in Fic55 mutants are partially rescued.

Expression of FicH375A.Scer\UAS under the control of Scer\GAL4repo fails to rescue the loss of the ON transients in Fic55 mutant electroretinograms. The defects in phototaxis seen in Fic55 mutants are also not rescued.
