UAS regulatory sequences drive expression of a DsRed(Unk) fluorescent protein. A genomic fragment containing an miRNA cluster (includes the mir-310, mir-311, mir-312 and mir-313 pre-miRNA hairpins) has been inserted into the DsRed(Unk) 3'UTR.
Clones expressing P{UAS-DsRed-mir-310+mir-311+mir-312+mir-313.L} (carries mir-310Scer\UAS.cluster.T:Disc\RFP, mir-311Scer\UAS.cluster.T:Disc\RFP, mir-312Scer\UAS.cluster.T:Disc\RFP and mir-313Scer\UAS.cluster.T:Disc\RFP) under the control of Scer\GAL4unspecified are difficult to recover in the wing imaginal disc. Expression of P{UAS-DsRed-mir-310+mir-311+mir-312+mir-313.L} under the control of Scer\GAL4Bx-MS1096 results in wings which are curved, small and folded.
Co-expression of BacA\p35Scer\UAS.cHa rescues the viability of clones in the wing disc expressing P{UAS-DsRed-mir-310+mir-311+mir-312+mir-313.L} (carries mir-310Scer\UAS.cluster.T:Disc\RFP, mir-311Scer\UAS.cluster.T:Disc\RFP, mir-312Scer\UAS.cluster.T:Disc\RFP and mir-313Scer\UAS.cluster.T:Disc\RFP) under the control of Scer\GAL4unspecified. The recovered clones are small, consisting of just one to three cells and appear to be delaminating from the epithelial plane.
