The neurotransmission across neuromuscular junctions in third instar larvae, assessed by excitatory junction potential amplitude and quantal content, is significantly impaired in Rim103/RimEx73 transheterozygotes, but not in Rim103 heterozygotes, as compared to controls.

Homozygous and RimEx73/Df(3R)ED5785 larvae show no gross morphological defects at the neuromuscular junction. Mutant boutons at the RimEx73/Df(3R)ED5785 neuroumuscular junction have normal active zone length, T-bar width and number of T-bars per active zone. There is a small decrease in the number of T-bars per bouton perimeter and in the number of active zones per bouton perimeter.

The average evoked excitatory junction current (EJC) amplitude at the neuromuscular junction is reduced by approximately 75% in RimEx73/Df(3R)ED5785 larvae compared to wild type. The average amplitude and frequency of the spontaneous mEJC is not significantly different from normal. There is a reduction in quantal content compared to wild type.

Cumulative amplitude analysis shows that the size of the readily releasable pool of vesicles at the neuromuscular junction is reduced in RimEx73/Df(3R)ED5785 larvae compared to wild type.

Expression of RimScer\UAS.cGa under the control of Scer\GAL4VGlut.PD partially rescues the reduction of the amplitude of the evoked excitatory junction current which is seen at the neuromuscular junction in RimEx73/Df(3R)ED5785 larvae.
