UAS regulatory sequences drive expression of a mutant form of Hsap\SERPINI1 in which the histidine at amino acid 338 has been replaced by arginine. The P1 and P1' amino acids (located in the reactive centre loop and forming the pseudosubstrate for the target proteinase) have been changed from Met-Arg to Pro-Pro. The construct includes the human signal secretion peptide.
The median lifespan of flies expressing Hsap\SERPINI1H338R.P1-P1'.Scer\UAS under the control of Scer\GAL4elav-C155 is similar to controls. Locomotor function is normal at eclosion but deteriorates rapidly, with clear deficits in climbing at 30 days.
